I want to create a schedule with the following criteria: - Monday to friday, 08:00 to 16:30 How should that period definition look like?

So it can be used to present a availability report for some sensors for "our business hours".

businesshours half-hours schedule

The last update releases of PRTG include an option for schedules entitled "Edit Mode." If you set the same to "Use list of period definitions" you can define schedules with greater granularity (down to 1 minute).

The line breaks are not showing in your post, but they were being displayed in the ticket we received, so yes, that looks about right.
